ION224 is an investigational DGAT2 antisense inhibitor in development for the treatment of metabolic dysfunction-associated steatohepatitis (MASH). The drug met the primary study endpoint at both doses (120 mg and 90 mg) in a Phase 2 trial, demonstrating statistically significant liver histologic improvement. ION224 also achieved the key secondary endpoint of MASH resolution without worsening of fibrosis, as measured by biopsy. Furthermore, the drug was safe and well-tolerated in study participants, with no on-study deaths or treatment-related serious adverse events. MASH, previously referred to as non-alcoholic steatohepatitis (NASH), has no approved drugs. The U.S. Food and Drug Administration is reviewing a potential therapy from rival Madrigal Pharmaceuticals ( MDGL ), with a decision expected by Thursday.
